    Source / Purification

    Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues surrounding glycine 109 of human TRIP-4/ASC-1. Antibodies are purified by protein A and peptide affinity chromatography.

    Background

    TRIP-4 (thyroid hormone receptor interactor 4)/ASC-1 (activating signal cointegrator 1) is a widely expressed transcriptional coactivator of nuclear receptors (1-3). The protein contains a zinc finger motif that serves as a binding site for basal transcription factors, TATA-binding protein (TBP) and TFIIA as well as SRC-1, CBP/p300 and nuclear receptors. Subcellular localization of TRIP-4/ASC-1 may depend on the cellular context. While it is normally expressed in the nucleus, TRIP-4/ASC-1 was observed in the cytoplasm under serum starvation but was retained in the nucleus when expressed with CBP or SRC-1 (2). TRIP-4/ASC-1 was also found to complex with AP-1, SRF and NF-κB and may play an important role in transactivation by these proteins as well as mediate their cross-talk with nuclear receptors (3).
