    Background

    Aminoacyl-tRNA synthetases are highly conserved enzymes that catalyze the aminoacylation of tRNA by their cognate amino acid, allowing the incorporation of the correct amino acid into a growing polypeptide chain and ensuring translation accuracy.

    Mutations in the gene encoding valine aminoacyl-tRNA synthetase (VARS), also known as valyl-tRNA synthetase (ValRS), are associated with neurodevelopmental disorders (1). High VARS expression linked to poor prognosis has been reported in human cancer, making VARS a potential therapeutic target (2). In BRAF V600E melanoma, VARS has been shown to drive resistance to MAPK inhibitor therapy (3).
