Neuroinflammation is a condition observed in the central nervous system (CNS) in response to infection, toxic metabolites, traumatic injury, or autoimmunity.
Our protocol is as follows:REAGENTS:TPA (12-O-Tetradecanoylphorbol-13-Acetate) #4174Lipopolysaccharides (LPS) #14011DAY 1:
Cellular senescence is characterized by irreversible cell-cycle arrest in combination with a distinct secretory phenotype and expanded lysosomes in response to stress. Senescent cells accumulate in tissues during aging, and various markers of senescence

Western blotting using fluorescent-conjugated secondary antibodies - accurate quantification and two-color multiplexing using primary antibodies from different species.
Apoptosis is a type of programmed cell death that occurs normally throughout the lifespan and can also occur in response to harmful stimuli. Apoptotic cells are identified by their altered morphology, caspase activation, and the presence of damaged DNA.

The immune system is composed of tissues, cells, and molecules whose primary function is to detect, respond to, and eliminate pathogens and transformed cells.
The pathway induction from H202 treatment may be more robust than the LPS treatment. We have previously observed that buffer and temperature conditions at the lysis stage can affect basal levels of PARylation. Notably, lysis at RT yielded lower observable PAR than lysis on ice (4 deg C). Also, performing the initial lysis in a PDE assay buffer (available from several vendors) yielded higher observable PAR than lysis directly into SDS loading buffer with DTT. Lastly, cells overgrown in culture may also show higher basal levels of PAR.

Understanding how TLR signaling is activated by molecular patterns is relevant not only to their role in combating infection but also in cancer.
Pyroptosis is a type of programmed necrotic cell death activated by infections from bacteria, viruses, fungi or protozoa in the presence of PAMPs or DAMPs
