Which total AMPKα antibody do you suggest for my western blot experiment?

The choice of AMPKα total antibody depends on the subunit of interest and the secondary antibody to be used. Cell Signaling Technology (CST) has five AMPKα antibodies that detect total protein: 

  • AMPKα Antibody #2532
  • AMPKα (D5A2) Rabbit mAb #5831
  • AMPKα (D63G4) Rabbit mAb #5832
  • AMPKα (F6) Mouse mAb #2793
  • AMPKα (23A3) Rabbit mAb #2603
It is important to note that AMPKα (D63G4) Rabbit mAb #5832 has been shown to recognize only AMPKα1 and not AMPKα2.

A side-by-side comparison of these five AMPKα antibodies has been performed (data available upon request). It revealed that the mouse-derived antibody, AMPKα (F6) Mouse mAb #2793, produces the strongest signal in western blotting applications. Of the rabbit-derived AMPKα antibodies, #5831 and #5832 show the strongest signals for western blotting. The polyclonal antibody, #2532, shows the weakest signal for western blotting applications.
