Can I use the Cyclic AMP XP® Assay Kit #4339 with tissue samples?

We have not tested tissue samples in-house using the Cyclic AMP XP Assay Kit #4339, so although we cannot guarantee it will work, there is no reason it should not. We recommend loading more protein (neat, undiluted) as there tends to be less protein in tissue samples. Cyclic nucleotides may be metabolized quickly in tissue, because negative regulation can occur in the pathway when phosphodiesterases (PDEs) catalyze the hydrolysis of cAMP to adenosine-5'-monophosphate (5'-AMP). Therefore, it is important to rapidly freeze tissues after collection (e.g., using liquid nitrogen). Without performing stability testing on tissue samples in-house, we are unable to provide a definitive answer on whether samples will contain detectable levels of cAMP.
