Which method is better for purifying CUT&Tag DNA, DNA affinity columns or a phenol-chloroform DNA extraction using ethanol precipitation?

We recommend using DNA Purification Buffers and Spin Columns (ChIP, CUT&RUN, CUT&Tag) #14209 when purifying your CUT&Tag DNA. Although phenol-chloroform extraction followed by ethanol precipitation will recover DNA fragments of all sizes, CST DNA spin columns effectively recover DNA fragments ≥ 35 bp. Because tagmented genomic DNA fragments are ≥ 70 bp, CST DNA spin columns offer a simple, inexpensive, and robust method for purifying your CUT&Tag DNA fragments.

For your convenience, DNA purification buffers and spin columns are included in the CST CUT&Tag Assay Kit #77552.
