What washing conditions do you recommend for immunofluorescent (IF) experiments?

Washing should be performed in 1X PBS. We recommend three 5-minute washes after the primary and secondary incubation steps, but 3 quick rinses should work as well, as long as you are working with cells or thin sections. We do not recommend adding detergent to the washing buffer as the increased stringency may lead to reduced antibody binding.
