The antigenic sequence used to produce our ATF-6 (D4Z8V) Rabbit mAb #65880 is located at the C-terminus of ATF-6, so this product may recognize a C-terminal fragment. However, our testing has shown that #65880 does not detect a cleavage product after stimulating ER-stress with an extended tunicamycin treatment (i.e., 8 hours).

UBA Protein Domain

Resources

First, ensure blood is collected in a vacutainer tube that does not contain any anticoagulants such as sodium heparin or EDTA.
1. In a biosafety hood transfer the blood into an appropriately sized conical tube or 1.5mL Eppendorf tube. Let the blood sit at room temperature for a least 1 hour until the blood clots.
2. Spin the tube at 1400rpm for 5 minutes. The resulting supernatant is the serum. Since the blood has had time to clot, the serum should be free of blood cells, fibrinogen, and platelets. Be careful not to disturb the pellet when pipetting off the serum.
3. Mix one part serum with one part 2X SDS/DTT. Sonicate the lysate and then boil for 5 minutes.

We see two bands migrating at 180 and 220 kDa in both endogenous and over-expressed lysates. We suspect these are differentially glycosylated forms of ROBO2 (UniProt ID Q9HCK4; https://www.uniprot.org/uniprot/Q9HCK4#ptm_processing).

We offer 3 MitoTracker® products, MitoTracker® Green FM #9074, MitoTracker® Deep Red FM #8778 and MitoTracker® Red CMXRos #9082.   MitoTracker® Deep Red FM #8778 and MitoTracker® Red CMXRos #9082 are well retained after methanol fixation as outlined in our Product Usage Information.  MitoTracker® Green FM #9074, on the other hand, is only useful for live cell imaging as the dye is not well retained after fixation with aldehydes or alcohols.

We test all unconjugated antibodies for 1 week (7 days) at -20C, room temperature (ambient) and 37C, then assess each stability test sample by western blot compared to the -20C stock. If there is no change in the performance of the antibody at room temperature or 37C by western blot after the 1 week period, the antibody is considered stable and shipped at ambient temperature without ice packs. If the antibody performance changes in any way after this time period, the antibody will be shipped cold with ice packs. Please note that regardless of the shipping method, the antibody should be stored at -20C upon arrival.

X-Gal can be tricky to solubilize, therefore we recommend dissolving in DMF (#12767) as listed on the Senescence β-Galactosidase Staining Kit (#9860) datasheet. However, we have tested and verified that DMSO (#12611) can be used to successfully dissolve the X-Gal, and therefore we consider it to be a suitable replacement for DMF.
