The most sensitive antibody will depend on whether you are trying to detect Cas9 protein from Streptococcus pyogenes, Staphylococcus aureus, or Campylobacter jejuni. The Cas9 (7A9-3A3) Mouse mAb #14697 and Cas9 (E7M1H) XP Rabbit mAb #19526 are the most sensitive choices by western immunoblot for the detection of Cas9 from Streptococcus pyogenes; whereas, the Cas9 (S. aureus) (E4G3U) Rabbit mAb #51610 is the most sensitive choice by western immunoblot for detection of Cas9 from Staphylococcus aureus. The Cas9 (C. jejuni) Antibody #50561 is currently our only antibody for western immunoblot for detection of Cas9 from Campylobacter jejuni.

Our Cas9 (7A9-3A3) Mouse mAb #14697 was produced with a recombinant protein specific to the amino terminus of Cas9 from Streptococcus pyogenes (UniProt ID #Q99ZW2; https://www.uniprot.org/uniprot/Q99ZW2). The precise epitope has not been mapped. However, this antibody has been shown to detect nuclease-deficient Cas9 (dCas9) protein and Cas9 nickase (niCas9) protein (please see the western blot image on the product datasheet; https://media.cellsignal.com/pdf/14697.pdf). dCas9 is a D10A/H840A mutant and niCas9 is a D10A mutant.

Yes, you may freeze your cells and pool them for a later CUT&RUN experiment. However, we would suggest a light fixation using 0.1% formaldehyde for 2 min before freezing the cells, in case the protein-DNA association inside the nuclei changes during the freeze-thaw of the cells.
For antigenic targets where expression of the protein is very low or unknown, the use of recombinant proteins or exogenous expression in a surrogate cell line may be necessary for antibody validation.

The average diameter of each Concanavalin A bead particle is ~1.0µm while the size range of a cell is 0.1-100 µm (the most commonly used cell lines like HeLa, HepG2 and HEK293 all have a cell size > 10µm). Therefore, in most cases, a cell is coated by multiple beads. This has also been confirmed by inspection under a microscope.
