Our ELISA kits are compatible with our RIPA Buffer (10X) #9806, however, this buffer may not work for some targets. RIPA buffer is very stringent in nature and can prevent the antibody/antigen from binding. For optimal performance, we suggest using the buffer included in the kit to generate your extracts. If you are generating tissue lysates, RIPA Buffer will better solubilize the proteins in the tissue compared to our Cell Lysis Buffer (10X) #9803 or PathScan® Sandwich ELISA Lysis Buffer (1X) #7018.

SignalStar Multiplex IHC Kits & Reagents have not yet been validated for use with antibodies outside of our menu. We currently offer custom conjugation of antibodies in our IHC-P-validated portfolio. In addition, SignalStar mIHC is a non-destructive technology that gives scientists the ability to perform other imaging assays on the same tissue, such as Hematoxylin and Eosin (H&E). We do not observe any bias toward euchromatin or heterochromatin in our assays. In CUT&RUN, the target-specific antibody recruits the pAG-MNase and directs digestion to chromatin that is directly adjacent to the antibody, whether that be in euchromatin or heterochromatin. In this case, the active tethering of pAG-MNase to the chromatin allows for digestion to occur even in less accessible heterochromatin. Our CUT&RUN Assay Kit #86652 has been shown to work well with antibodies against active, accessible euchromatic histone modifications (see Tri-Methyl-Histone H3 (Lys4) (C42D8) Rabbit mAb #9751) and activating transcription factors (see NF-kB p65 [D14E12] XP® Rabbit mAb #8242…
